E3 ubiquitin ligases are proteins that transfer Ubiquitin from an E2 ubiquitin-conjugating enzyme to a target destined for degradation. They are one of the binding partners of PROTACs and Molecular glues.

Per (1), there are many subtypes of E3 ubiquitin ligases (three of which are listed below):

  1. RING: largest class, simultaneously binds E2-Ub and target
  2. HCET: Transfer Ub to itself and then moves it to target
  3. RBR: Hybrid strategy
